Haemoglobin Estimation of blood donors (specific gravity method)


Test procedure:

1.    Dispense 30 ml of copper Sulphate solution into appropriate labeled, clean, dry tube or bottle. Change the solution daily or after 25 tests and be sure that the solution is properly mixed before use.
2.    Clean the finger tip with antiseptic and allow it to dry.
3.     Sterile lancet or needle is used for prick and there should be free flow of blood.
4.    The drop of blood is either collected in capillary tube or pipette and allows falling gently from a height of 1 can above the surface of the copper sulphate solution.
5.    The drop get enclosed in a sac of copper proteinate which prevents any change in spe. Gr. For about 15 seconds. Howeverifthe drop sinis Within 15 seconds Hb  of the blood is 12.5g/dl or above.



Sources of error

1.    Taking first drop of blood from the fingerprick
2.     Squeezing the finger because the blood not flowing freely
3.     Dirty pipette

4.     Chip of delivering end of pipette.
