Compatibility tests

Maior & minor compatibility test for IgM antibodies(Saline)

1.    Label 2 tubes as Major & minor
2.    Put 2 drops of recipient's serum in tube M’
3.     Put 2 drops of donor serum in tube *m'
4.     Add 1 drop of 2-4% saline suspension of donor cell to tube M’
5.     Add 1 drop of 2-4% saline suspension of recipient cell to tube 'm'
6.    Mix well and incubate for 5-10 min. (spin method) or incubate for 30-60 min. (sedimentation method) at RT.
7.    Centrifuge at 1000 rpm for 1 min.
8.    Read the result observe for haemaolysis and agglutination.
9.    Negative result should be confirmed under microscope Interpretation: Agglutination or haemolysis indicates a positive result (incompatible)
Anti-globulin compatibility test for IgG antibodies (IAT)

1.    Use the same tubes of saline compatibility test
2.   Incubate the tubes for30-60 minutes at 37°C.
3.   Centrifuge at 1000 rpm for 1 min. checkfor haemolysis or agglutination
4.   If there is no haemolysis/agglutination, wash the cells 3 times with normal saline.
5.   Perform IAT test:
a.      Add 2 drops of AHG serum to washed cells
b.     Centrifuge at 1000 rpm for 1 minute
c.     See for agglutination


Interpretation:

 Haemolysis or agglutinationatany stage indicate incompatibility
